Novel monovalent and multivalent recombinant proteins of Borrelia burgdorferi sensu lato as potential antigens for the detection of specific antibodies in the sera of Lyme patients

The aim of the present project is the development of novel monovalent and multivalent Borrelia burgdorferi sensu lato (sl) recombinant proteins and the determination of their antigenic properties. Diagnosis of Lyme borreliosis (LB), an infection caused by spirochetes of the Borrelia burgdorferi sl species complex, is mainly based on clinical symptoms supported with serology and is often misdiagnosed in areas of endemicity. In Central and Eastern Europe, LB is most often caused by B. burgdorferi sensu stricto (ss), B. garinii, B. afzelii, although the remaining European species, B. valaisiana, B. lusitaniae, B. bissettii and B. spielmanii, have also been isolated from body fluids and tissues of patients with symptoms of borreliosis. The complexity of the antigenic composition among the Borrelia genospecies and differential expression of proteins in host and vector (temporal and spatial antigenic variability) has posed challenges for the serodiagnosis of LB. Therefore, despite many efforts, the correct diagnosis of this disease still faces many difficulties. For this reason, it is extremely important to conduct basic research aimed at selecting, receiving, and testing the antigenic properties of novel recombinant proteins with potential diagnostic usefulness.
