Zastosowanie modelu glonowego do określenia mitotoksyczności niesteroidowych leków przeciwzapalnych na przykładzie diklofenaku

Diclofenac (DF), belonging to the non-steroidal anti-inflammatory drugs (NSAIDs), is one of the pharmaceuticals on the list of priority hazardous substances for environment. Pharmaceuticals, designed to have a specific therapeutic effect on humans and animals, could have significant toxic effect on non-target organisms living in polluted areas. It was shown that in animal cells DF disrupt mitochondrion functioning due to uncoupling of oxidative phosphorylation by the opening of membrane permeability transition pore in the mitochondria, which leads to the loss of the transmembrane potential required for synthesis of ATP. DF also disturb Ca2+ homeostasis because it blocks NMDA-receptor, the glutamate-gated channel responsible for Ca2+ transport to the cell. Analogs of the above channels were found in plants and algae which suggests that DF would affect them in a similar way. Because Ca2+ is necessary to keep mitochondrion and chloroplast (energetic centers of plant cells) operate, it cannot be excluded that changes in intracellular Ca2+concentration, leading to disruption of cell bioenergetics, could be one of the reasons of DF phytotoxicity. In few reported studies concerning pharmaceuticals effect on plants, photosynthesis disruption was shown but respiratory processes were not investigated in detail. Thus, the aim of the project is to verify the hypothesis that one of the causes of DF phytotoxicity is the disorder of plant mitochondrion functioning, based on the model green algae Chlamydomonas reinhardtii.
