Innovative stent coating with polyphenol-rapamycin complex with improved drug-eluting properties to target in-stent-restenosis

PoRaCoat aims to develop an innovative drug-eluting stent coating containing the drug rapamycin incorporated into polifenol-Cu2+ complex, with known anti-inflammatory and anti-mitotic actions and a conductive polymer matrix based on poly(3,4-ethylenedioxythiophene) (PEDOT), being a very promising coating material for biomedical applications due to its high biocompatibility. The aim is to improve the release of rapamycin during degradation in a concentration-dependent manner to achieve beneficial effects to promote re-endothelialization while inhibiting a new thickening of the vessel wall due to the adhesion and proliferation of smooth muscle cells. After hemo- and cytocompatibility testing, effects on the function of endothelial and smooth muscle cells will be analyzed. Finally, a stent coating demonstrator will be developed that will be undergoing preclinical testing using cells from patients with PAD.The consortium of five project partners, the Fraunhofer Institute for Ceramic Technologies and Systems IKTS and University Hospital Dresden, Division of Vascular and Endovascular Surgery from Germany, the Gdańsk University of Technology and the Balton company from Poland and the University POLITEHNICA of Bucharest from Romania, possesses all necessary competences and experience to perform the proposed study. The project will allow for the performance of important research, the results of which will be gradually published in peer-reviewed JCR journals. The developed technology will be patented. The results of the project will be presented at different international and national conferences. Moreover, ongoing projects will allow for the purchase of new equipment and laboratory accessories.
