The influence of reactions conditions on aggregation of dyes used in biochemistry

Abstract

Self-association of dyes and related substances is a very important phenomenon in many fields of applied chemistry. It is also fundamental model reaction of many kinds of molecular interactions such as the micelle formation of amphiphilic substances and the binding of small organic molecules to macromolecules. In spite of many studies concerning the dimerization equilibria of dyes and related substances, the mechanism seems not to be full recognized because the experimental conditions such as solvent coexisting salt and its concentration are different from study to study.The formation of the aggregates can be observed by the change in the electronic absorption in UV-VIS Spectroscopy as well as entalphy change in isothermal titration calorimetry (ITC). In the analysis of the absorption data, principal component analysis (PCA) and some optymalization methods have been applied to determine dimerization constants. Using of ITC permits direct determination of enthalpy changes associated with reactions. To analyze termodynamic process of deagreggation we compile special algorithm which assume that basic step of agregation is dimerization (which is in the agreement with spectroscopic data).In our work the spectroscopy and microcalorimetry technique have been used to analyze aggregation process of typical dyes used in biochemistry. We have compared different reaction conditions such as temperature, salt concentration and pH resulting in correlation between dimerization constant and analyzed parameters.
