Abstract
Obecnie reakcje PCR (ang. Polymerase Chain Reaction) wykazują bardzo szerokie zastosowanie w diagnostyce medycznej, biologii molekularnej czy inżynierii genetycznej. Efektywność tych reakcji rozumiana jako wydajność i wierność przeprowadzonej amplifikacji jest nieodłącznie związana ze stosowaną polimerazą DNA i warunkami prowadzenia reakcji PCR. Aby sprostać wymaganiom stawianym przez nowoczesne metody diagnostyczne oraz współczesną inżynierię genetyczną konieczne jest poszukiwanie polimeraz DNA o coraz lepszych właściwościach. Dotychczas wprowadzane modyfikacje amplifikacji DNA to przede wszystkim stosowanie udoskonalonych buforów, wzmacniaczy reakcji PCR, wprowadzanie mutacji w polimerazach DNA lub konstrukcje polimeraz chimerycznych. Coraz popularniejsze stają się również modyfikacje znanych już polimeraz DNA poprzez tworzenie ich fuzji z białkami mogącymi poprawić ich użyteczność. Podstawowym założeniem niniejszej pracy było otrzymanie nowej, lepszej polimerazy DNA jako wynik fuzji polimerazy DNA z białkiem wiążącymi jedno- lub dwuniciowe DNA i zbadanie jej nowych właściwości pod względem procesywności, termostabilności, powinowactwa do matrycy DNA, jak również odporności na inhibitory występujące w próbkach klinicznych (np. we krwi). Modyfikacjom została poddana najbardziej popularna w diagnostyce molekularnej polimeraza DNA z Thermus aquaticus z delecją reszt aminokwasowych tworzących domenę odpowiedzialną za aktywność egzonukleolityczną 5’→3’ (TaqStoffel). Natomiast białkami, które zastosowano do fuzji były białka mające naturalną zdolność do wiązania się z DNA: RB69SSB, TtePriB i NeqSSB oraz domena DBD ligazy P. furiosus. Cechy nowych polimeraz DNA porównano z polimerazą DNA TaqStoffel. Uzyskano polimerazy o ulepszonych podstawowych cechach, bardziej przydatnych do pracy z problematycznymi matrycami (długie i bogate w pary GC) oraz z próbkami, którym mogą towarzyszyć inhibitory z krwi (laktoferyna, heparyna). Ponadto wykazano wzrost termostabilności, procesywności i czułości fuzyjnych polimeraz DNA zwłaszcza w przypadku fuzji z białkiem SSB pochodzącym z Nanoarchaeum equitans (NeqSSB) oraz domeną DBD ligazy Pyrococcus furiosus.Zoptymalizowane i przeprowadzone doświadczenia pozwoliły również na wyznaczenie istotnych cech białek użytych w fuzji z polimerazą DNA (co może być istotne w racjonalnym projektowaniu innych fuzyjnych polimeraz DNA) oraz sposobu badania ich właściwości.
