Kinetics of cyclohexane removal by Candida subhashii and Fusarium solani

Description

Dataset presents cyclohexane biodegradation in gas phase using two species of microorganisms: Candida Subhashii and Fusarium solani. Biodegradation was carried out in sealed chambers with a capacity of 1000 ml. In each of them there are two discs inhabited with microorganisms, made of polyurethane foam, 80 mm in diameter and 20 mm thick.

An immobilization of Candida subhashii on the support materials was carried out in 500 ml sterile beakers containing single type of the substrate and 100 mL growth Sabouraud medium with 10% (v/v) inoculum, under orbital agitation at 100 rpm and at 24°C. After 24 h, 50% of the volume of medium was replaced with the same volume of the minimal medium: Na2HPO4•2H2O (15.2 g/dm-3), KH2PO4 (3 g/dm-3), NaCl (0.5 g/dm-3) and NH4Cl (1 g/dm-3). The beakers with appropriate contents were shaken throughout the whole immobilization. The next day, all medium was poured out, replacing it with the minimal medium. The procedure was repeated after one day with the difference that the medium spilled was replaced with the Sabouraud medium. After the same time interval the support materials were completely colonized 
The immobilization of Fusarium solani on the support particles was carried out on Petri plates (10 cm) with Sabourand Agar medium. Using inoculation loop seed individual fungal colonies on plates. Incubate the plates at 24°C for 3-4 days. After this time, were placed the the support materials in the plates. Incubate the plates at 24°C for 5-6 days. After 8-10 days from the beginning, the polyurethane foam material was completely colonized.
In the next step, an appropriate amount of cyclohexane and toluene (in case of a mixture) were injected into each chamber. The amounts of the substances were selected so that the initial concentration of cyclohexane was about 400-500 ppm v/v, and of toluene 200-300 ppm v/v (in gas phase).
The cycloehxane concentrations were determined using the gas chromatography technique. The obtained results of the cyclohexane concentrations are presented in four files:
1. Candida_subhashii_cyclohexane - presents cyclohexane concentration changes in the chamber filled with a one-component mixture (only cyclohexane)
2. Candida_subhashii_mixture - presents cyclohexane concentration changes in the chamber filled with a multi-component mixture (cyclohexane and toluene)
3. Fusarium_solani_cyclohexane - presents cyclohexane concentration changes in the chamber filled with a one-component mixture (only cyclohexane)
4. Fusarium_solani_mixture - presents cyclohexane concentration changes in the chamber filled with a multi-component mixture (cyclohexane and toluene)
