Determination of high-intensity sweeteners in beverages by high-performance liquid chromatography with mass spectrometry detection.

Abstract

The objective of the present study was to measure the concentration of nine high intensity sweeteners (acesulfame-K, aspartame, alitame, cyclamate, dulcin, neohesperidin dihydrochalcon, neotame, saccharin and sucralose) in different types of beverages available on the polish market. The proposed methodology reported here consists of a extraction with buffer composed of formic acid and N,N-diisopropylethylamine (pH 4.5) followed by isolation of sweeteners with use of solid-phase extraction using Strata-X 33µm Polymeric SPE column. The chromatographic separation of the sweeteners was achieved using C18 column and a gradient elution. MS detection was performed in a time-scheduled SIM mode, using electrospray interface operating in the negative ion mode.Several validation parameters, such as linearity, limits of detection and quantification, accuracy and precision were evaluated. Calibration curves were linear within a studied range of concentrations with correlation coefficients over 0.999 for six investigated sweeteners (CYC, ASP, ALI, DUL, NHDC, NEO). Three compounds (ACS-K, SAC, SCL) gave non-linear response in the investigated concentration range. The method detection limits (MDLs) were below 0.25 µg/ml, whereas the limits of quantification (MQLs) were below 2.5 µg/ml. The recoveries at the tested concentrations (50, 100 and 125 % of MUD value) for all sweeteners were 80.9-106.7 %, with RSDs 5.3 %. regardless of the spiking level. The intra-day precision was between 0.2-4.7 % and the inter-day precision was between 0.4-6.3 %.
