Możliwości zastosowań, potencjalne źródła oraz ewolucja technologiczna sposobu otrzymywania trehalozy ze szczególnym uwzględnieniem enzymu syntazy trehalozy [EC 5.4.99.16]
Abstract
Jedna z wiodących gałęzi współczesnej biotechnologii opiera się na produkcji białek – przede wszystkim enzymów. Są one powszechnie wykorzystywane: od przemysłu chemicznego poprzez przemysł spożywczy aż po medycynę i życie codzienne (czy ktoś wyobraża sobie w dzisiejszych czasach proszek do prania „bez enzymu”). Przed rozwojem nowoczesnych metod biologii molekularnej i inżynierii genetycznej pozyskiwano interesujące substancje z tkanek organizmów żywych. Ze względu na ciągle rosnące zapotrzebowanie, ta droga izolacji/produkcji okazała się niewystarczająca. Zastosowanie technik opartych o modyfikacje DNA umożliwiło masową produkcję prowadzoną w specjalnych systemach ekspresyjnych. Autor w rozdziale opisuje przypadek zmiany sposobu pozyskiwania specyficznego cukru o wielu unikalnych właściwościach jakim jest trehaloza. W bardzo szybkim czasie okazało się, że „klasyczna” metoda izolacji tego cukru z roślin, a następnie także już w procesie biotechnologicznym z zastosowaniem drożdży, nie jest wystarczająco wydajna. Do tego jest obarczona wysokimi kosztami i dużym poziomem skomplikowania. Zastosowanie podejścia rodem z kolejnej fali rozwoju procesów biotechnologicznych pozwoliło ostatecznie wprowadzić ją na rynek w wystarczającej ilości, a nawet w wyniku sprzężenia zwrotnego (jak i z powodu unikalnych właściwości trehalozy) stale zwiększać na nią zapotrzebowanie. W opracowaniu można znaleźć syntetyczne opis potencjalnych źródeł enzymów biosyntetyzujących trehalozę, krótki opis szlaków biochemicznych syntezy trehalozy w organizmach żywych, charakterystykę enzymu o największym potencjale aplikacyjnym (według autora – syntazy trehalozy), potencjalne i praktyczne zastosowania samej trehalozy jak i opis zmian w technologii wytwarzania tego cukru na przestrzeni lat, a także pozostałe do rozwiązania problemy
