Njwyższą wydajność syntezy uzyskuje się przy pH 6-9 i 10-30% stężenia wody. Podobnie jak dla mezofilnych beta-galaktozydaz enzym antarktyczny syntetyzował najwydajniej kiedy reakcja przebiega w mieszaninie buforu i rozpuszczalnika organiczego (poniżej 50%, v/v).
