Antifungal Effect of Penicillamine Due to the Selective Targeting of L-Homoserine O-Acetyltransferase
PublikacjaDue to the apparent similarity of fungal and mammalian metabolic pathways, the number of established antifungal targets is low, and the identification of novel ones is highly desirable. The results of our studies, presented in this work, indicate that the fungal biosynthetic pathway of L-methionine, an amino acid essential for humans, seems to be an attractive perspective. The MET2 gene from Candida albicans encoding L-homoserine...

L-Homoserine O-acetyltransferase (CaMet2p) activity and inhibition
Dane BadawczeThe dataset contains the description of methodology and data files with the results obtained spectrophotometrically at 412 nm to determine the rate of fungal enzyme activity. The goal of the study was to obtain conditions optimal for L-homoserine O-acetyltransferase (Met2p) activity as well as evaluation of inhibitory potential of commercially available...

Antifungal activity of L-homoserine O-acetyltransferase (CaMet2p) inhibitors.
Dane BadawczeThe dataset contains a set of optical densities at 600 nm measured at microplate reader to determine the rate of fungal growth in the presence of Candida albicans homoserine O-acetyltransferase inhibitors. Antifungal activities were determined against several strains from Candida spp. and others by the modified M27-A3 method specified by the CLSI. We...

Activity assay of O-Acetyl-L-homoserine sulfhydrylase (CaMet15p).
Dane BadawczeThe study aimed to obtain conditions optimal for the activity of O-acetyl-L-homoserine sulfhydrylase (CaMet15p) and a method suitable for its measurement. The selection of appropriate reaction conditions included the identification of the reaction buffer and its pH, substrate and enzyme concentrations. The activity of CaMet15p was measured by the detection...

Overproduction of homoserine O-acetyltransferase (CaMet2p) native and His-tag versions.
Dane BadawczeEnzyme of fungal L-methionine biosynthetic pathway: homoserine O-acetyltransferase (Met2p) could be exploited as molecular target for antifungal chemotherapy. The goal of the study was to obtain conditions optimal for protein production with the use of prokaryotic cells. The constructed expression plasmids, designed in three different versions: encoding...

Identification and cloning of C. albicans SC5314 genes encoding L-methionine biosynthetic pathway enzymes.
Dane BadawczeEnzymes of fungal L-methionine biosynthetic pathway: homoserine O-acetyltransferase (Met2p), O-acetylhomoserine sulfhydrylase (Met15p) and cystathionine-γ-synthase (Str2p) could be exploited as molecular targets for antifungal chemotherapy. The goal of the study was to identify and clone genes encoding mentioned above enzymes. MET2, MET15 and STR2 genes...

Inserts amplification for knockout cassette construction
Dane BadawczeThe goal of the study was to obtain optimal conditions for amplification of upstream and downstream genes fragments and introduce a cleavage site for restriction enzymes into them thanks to which it will be possible to clone fragments into a plasmid containing elements of the knockout cassette. The constructed cassettes will enable the removal of the...

Antifungal dipeptides incorporating an inhibitor of homoserine dehydrogenase
PublikacjaThe antifungal activity of 5‐hydroxy‐4‐oxo‐L‐norvaline (HONV), exhibited under conditions mimicking human serum, may be improved upon incorporation of this amino acid into a dipeptide structure. Several HONV‐containing dipeptides inhibited growth of human pathogenic yeasts of the Candida genus in the RPMI‐1640 medium, with minimal inhibitory concentration values in the 32 to 64 μg mL−1 range. This activity was not affected by multidrug...

The bioactivity of processed garlic (Allium sativum L.) as shown in vitro and in vivo studies on rats

Three-dimensional Weyl topology in one-dimensional photonic structures
PublikacjaTopological features, in particular distinct band intersections known as nodal rings, usually requiring three-dimensional structures, have now been demonstrated experimentally in an elegantly simple one-dimensional photonic crystal.

The application of Monod equation to denitrification kinetics description in the moving bed biofilm reactor (MBBR)
PublikacjaIn this paper, the kinetic constants Vmax and KCOD occurring in the Monod equation, which describe the denitrification process in the moving bed, are determined. For this purpose, a laboratory moving bed biofilm reactor (MBBR) was used. The filling of the reactor consisted of EvU Perl carriers. The experiment was carried out with an excess of nitrate, and denitrification rate was dependent on the concentration of external organic...

KONSTRUKCJA AUKSOTROFICZNEGO WZGLĘDEM L-LIZYNY SZCZEPU CANDIDA ALBICANS

Syntheses and crystal structures of lithium derivatives of diphosphanes R2P-P(SiMe3)Li·3L, R = Ph, iPr and iPr2N, L = THF or DME
W reakcji BuLi z Ph2P-P(SiMe3)2 powstają znaczne ilości produktów ubocznych: Ph2P-PPh2 i LiP(SiMe3)2.

Double ZIF-L structures with exceptional CO2 capacity
PublikacjaCarbon dioxide emission is an emerging problem nowadays and new methods are designed for its control. In this article, a report on the formation of zeolitic imidazole framework with an apparent double leaf-like morphology DZIF-L is given. The special structure of the materials prevents from aggregation of the particles providing remarkable CO2 capacity. At 0.1 MPa the CO2 uptake was 2.99 ± 0.06 mmol/g. The capacity of synthesized...

N-Alkyl Derivatives of L-Glutamine As Inhibitors of Glutamine - Utilizing Enzymes
PublikacjaA general facile method to synthesize the N-gamma-alkyl and N-gamma,N-gamma-dialkyl derivatives of L-glutamine (1a-d) from L-glutamic acid as a starting substrate is presented. The obtained compounds are shown to inhibit three diferent glutamine-utilizing enzymes, namely: glutaminase, gamma-glutamyl transpeptidase, and glucosamine-6-phosphate synthase, with inhibitory constants within the milimolar range.
