Effect of Maximal Breath Hold Apnea on Pial Artery Pulsation and Subarachnoid Width in Human
PublikacjaLittle is known about intracranial pressure (ICP)-cerebral hemodynamic interplay during maximal breath-hold apnea. A recently developed method based on near-infrared transillumination/backscattering sounding (NIR-T/BSS) noninvasively measures changes in pial artery pulsation (cc-TQ) as well as subarachnoid width (sas-TQ) in humans. Changes in sas-TQ correlate with ICP to a considerable extent, whereas cc-TQ reflects the functional...

Immunophenotyping and transcriptional profiling of in vitro cultured human adipose tissue derived stem cells
PublikacjaAdipose-derived stem cells (ASCs) have become an important research model in regenerative medicine. However, there are controversies regarding the impact of prolonged cell culture on the ASCs phenotype and their differentiation potential. Hence, we studied 10 clinical ASCs replicates from plastic and oncological surgery patients, in six-passage FBS supplemented cultures. We quantified basic mesenchymal cell surface marker transcripts...

Wavelet transform analysis to assess oscillations in pial artery pulsation at the human cardiac frequency
PublikacjaPial artery adjustments to changes in blood pressure (BP)may last only seconds in humans. Using a novelmethod called near-infrared transillumination backscattering sounding (NIR-T/BSS) that allows for the non-invasive measurement of pial artery pulsation (cc-TQ) in humans, we aimed to assess the relationship between spontaneous oscillations in BP and cc-TQ at frequencies between 0.5 Hz and 5 Hz. We hypothesized that analysis of...

Human UDP-Glucuronosyltransferases: Effects of altered expression in breast and pancreatic cancer cell lines.
PublikacjaIncreased aerobic glycolysis and de novo lipid biosynthesis are common characteristics of invasive cancers. UDP-glucuronosyltransferases (UGTs) are phase II drug metabolizing enzymes that in normal cells possess the ability to glucuronidate these lipids and speed their excretion; however, de-regulation of these enzymes in cancer cells can lead to an accumulation of bioactive lipids, which further fuels cancer progression. We hypothesize...

Induction of apoptosis by triazoloacridinone derivative - C-1533 in HL60 and MOLT4 human leukemia cells.
PublikacjaPrzeprowadzone badania miały na celu określenie zdolności triazoloakrydonu C-1533 do indukcji śmierci komórek nowotworowych MOLT4 i HL60 na drodze apoptozy. Badanie zjawiska apoptozy przeprowadzono z wykorzystaniem techniki cytofluorymetrii przepływowej, mikroskopii fluorescencyjnej i elektroforezy agarozowej. Badania wykazały, że zarówno w komórkach MOLT4 jak i HL60, spadek mitochondrialnego potencjału błonowego oraz translokacja...

The effect of new non-cross resistant antitumor agents on the energy state of human erythrocytes

Design, synthesis and high antitumor potential of new unsymmetrical bisacridine derivatives towards human solid tumors, specifically pancreatic cancers and their unique ability to stabilize DNA G-quadruplexes
PublikacjaNew promising unsymmetrical bisacridine derivatives (UAs), have been developed. Three groupsincluding 36 compounds were synthesized by the condensation of 4-nitro or 4-methylacridinone, imi-dazoacridinone and triazoloacridinone derivatives with 1-nitroacridine compounds linked with anaminoalkyl chain. Cytotoxicity screening revealed the high potency of these compounds against severaltumor cell lines. Particularly, imidazoacridinone-1-nitroacridine...

PARP inhibition potentiates the cytotoxic activity of C-1305, a selective inhibitor of topoisomerase II, in human BRCA1-positive breast cancer cells
PublikacjaTwo cellular proteins encoded by the breast and ovarian cancer type 1 susceptibility (BRCA1 and BRCA2) tumor suppressor genes are essential for DNA integrity and the maintenance of genomic stability.Approximately 5-10% of breast and ovarian cancers result from inherited alterations or mutations in these genes.Remarkably, BRCA1/BRCA2-deficient cells are hypersensitive to selective inhibition of poly(ADPribose) polymerase 1 (PARP-1),...

Discriminating macromolecular interactions based on an impedimetric fingerprint supported by multivariate data analysis for rapid and label-free Escherichia coli recognition in human urine
PublikacjaThis manuscript presents a novel approach to address the challenges of electrode fouling and highly complex electrode nanoarchitecture, which are primary concerns for biosensors operating in real environments. The proposed approach utilizes multiparametric impedance discriminant analysis (MIDA) to obtain a fingerprint of the macromolecular interactions on flat glassy carbon surfaces, achieved through self-organized, drop-cast,...

A new dilution-enrichment sample preparation strategy for expanded metabolome monitoring of human breast milk that overcomes the simultaneous presence of low- and high-abundance lipid species
PublikacjaThe complex nature of human breast milk (HBM) makes samples difficult to analyze, requiring several extraction techniques and analytical platforms to obtain high metabolome coverage. In this work, we combined liquid-liquid extraction (LLE) and solid-phase extraction (SPE) techniques to prepare HBM samples to overcome the challenge of low- and high-abundance lipid species, enabling the semiquantitative analysis of total HBM lipids...

Intelligence Augmentation and Amplification: Approaches, Tools, and Case Studies
PublikacjaMost experts agree that truly intelligent artificial system is yet to be developed. The main issue that still remains a challenge is imposing trust and explainability into such systems. However, is full replication of human intelligence really desirable key aim in intelligence related technology and research? This is where the concept of augmented intelligence comes into play. It is an alternative conceptualization of artificial...

Modulation of CYP1A1, CYP1A2 and CYP1B1 expression by cabbage juices and indoles in human breast cell lines
PublikacjaEpidemiological studies have shown that consumption of cab- bage and sauerkraut is connected with significant reduction of breast cancer incidences. Estrogens are considered a major breast cancer risk factor and their metabolism by P450 enzymes substan- tially contributes to carcinogenic activity. The aim of this study was to investigate the effect of cabbage and sauerkraut juices of differ- ent origin on the expression profile...
