PublicationTermofilna bakteria Thermus ruber wytwarza syntazę trehalozy użyteczną do jednoetapowego przekształcania maltozy w trehalozę. Enzym ten wykazuje największą aktywność w temperaturze 65 C przy pH 6,5. Największą wydajność wytwarzania enzymu osiągano podczas hodowli drobnoustroju w temp. 55 stC w podłożu zawierającym 0,5% peptonu,0,1% ekstraktu drożdżowego oraz 0,% maltozy lub skrobi.

PublicationBiałka SSB są niezbędne do życia każdemu organizmowi. W tej pracy prezentujemy wyniki otrzymania i oczyszczania białek SSB Thermus thermophilus i T. aquaticus. Dotąd były znane białka SSB jedynie o budowie homotetrameru lub heterotrimeru. Białka SSB Thermus thermophilus i T. aquaticus stanowią nową klasę białek SSB - białek homodimerycznych. Ponadto dowiedziono, iż białka te są użyteczne w reakcji PCR.

PublicationW tej publikacji prezentujemy zastosowanie białka SSB-podobnego Thermus aquaticus (TaqSSB) w reakcji multiplex PCR w celu identyfikacji markerów ludzkiego Y-STR. Wykorzystanie termostabilnego białka TaqSSB chroni lub redukuje tworzenie przez startery struktur dimerycznych, które powodują hamowanie hybrydyzacji starterów do badanego DNA i redukują ilość starterów dostępnych w reakcji PCR.
